DSIP

Delta Sleep-Inducing Peptide — The Original Sleep-Architecture Research Compound

DSIP (Delta Sleep-Inducing Peptide) is a naturally occurring nonapeptide first isolated in 1974 from the cerebral venous blood of rabbits in induced slow-wave sleep. Unlike sedative compounds, DSIP has been studied as a neuromodulator that appears to influence sleep architecture — particularly delta-wave (slow-wave) sleep — as well as stress-hormone regulation, thermoregulation and pain thresholds. It remains one of the longest-studied endogenous peptides in sleep and neuroendocrine research.

  • Endogenous nonapeptide isolated in 1974 from slow-wave-sleep brain dialysate
  • Studied for delta-wave sleep architecture, cortisol regulation and pain thresholds
  • Crosses the blood-brain barrier; five decades of published neuroendocrine literature

Isolation and characterisation of DSIP

A humoral factor from the cerebral venous blood of rabbits in slow-wave sleep induced delta-wave sleep when transfused into recipient animals; the nonapeptide was sequenced and named DSIP.

Proceedings of the National Academy of Sciences · 1977

DSIP in chronic insomnia patients

Administration was associated with reduced sleep-onset latency and improved sleep efficiency in a small clinical series of chronic insomniacs.

European Neurology · 1984

Stress-axis and ACTH modulation

DSIP administration attenuated stress-induced corticotropin and corticosterone elevations in rodent models.

Neuroendocrinology · 1986

Analgesic and withdrawal models

DSIP raised pain thresholds and reduced withdrawal symptom severity in opiate- and alcohol-withdrawal animal and clinical pilot models.

Progress in Neuro-Psychopharmacology · 1984

Endogenous DSIP-like immunoreactivity in humans

DSIP-like immunoreactivity was detected in human plasma, cerebrospinal fluid and breast milk, with circadian variation in plasma levels.

From the field

What researchers report

Aggregated, anonymized observations from laboratories ordering this compound. Anecdotal and non-clinical.

Overwhelmingly selected for evening-administration study designs to align with the natural delta-sleep window.

Researchers frequently describe the effect profile in models as non-sedative — sleep architecture shifts rather than sedation onset.

Commonly run alongside GH-secretagogue arms because deep-sleep GH pulses are a shared endpoint.

Short plasma half-life is the most-cited design constraint; timing precision is emphasised in protocols.

Background

Research background

DSIP was isolated in 1974 by Monnier and Schoenenberger from dialysate of rabbits in electrically induced slow-wave sleep.

It crosses the blood-brain barrier readily despite its small size, and has been detected endogenously in human plasma, CSF and breast milk.

Research interest has broadened from sleep into stress-hormone regulation, chronic pain models and withdrawal syndromes.

Schoenenberger & Monnier (PNAS, 1977): DSIP isolated as the humoral factor associated with delta-wave sleep induction.

Answers

DSIP — frequently asked

Is DSIP a sedative?

No — the published literature describes it as a neuromodulator that influences sleep architecture, not a GABAergic sedative.

Is DSIP naturally occurring?

Yes. DSIP-like immunoreactivity has been detected endogenously in human plasma, cerebrospinal fluid and breast milk.

Why is the human evidence limited?

Most DSIP work dates from the 1970s–1990s and consists of small clinical series and animal models; it has never gone through modern large-scale trials.

What purity do you guarantee?

≥99% by HPLC with a batch-matched third-party COA.

How fast does it ship?

Orders placed before 2pm ET ship the same business day from Toronto with tracked Canada-wide delivery, typically arriving in 1–3 business days. Cold-pack packaging is used on every shipment.

What if the purity doesn't match the COA?

Every batch is HPLC and mass-spec verified by an ISO 17025 accredited third-party lab, and the batch-matched COA ships with your order. If an independent test comes back below the stated purity, we replace the order or refund it in full.

Is this legal to purchase in Canada?

Yes — these compounds are sold strictly as in-vitro research chemicals for laboratory use. They are not for human or veterinary use, and it is the purchaser's responsibility to comply with applicable federal and provincial regulations.

Ready to add DSIP to your research?

≥99% HPLC-verified, endotoxin and heavy metals tested, batch-matched COA included. Ships same business day from Toronto.
